Simple question...

If I have a website http://www.landscapers.com and the domain http://www.seanslandscaping.com redirects to it, can I do SEO for http://www.seanslandscaping.com and get that domain ranked or is that not possible?

Redirecting 301 a domain name means passing all the old links value to the new domain name..
So despite keep on working the old domain name you should start focus on building backlinks for a new domain name..

301 redirect is a domain name and it's mean linking old links to new website domain name. The new website using the links of old links. You can also generate new natural backlinks to your sites, It's much useful for your new website.

My two cents is that 301's work best when they re-direct to an inner page of your site rather than the homepage. Get a load of links to the site that you are redirecting and then let it be for a few weeks. The thing is with 301s is that they have a limited shelf life in terms of worth to your money site but again, this is just my personal experience.
