I want to know when I create a backlink let's say I create backlink from Article directory and it's also show in google indexing is this count in my backlink profile or not and does in this case Dofollow and Nofollow matters.Share your expert opinions....

Your Senior is right Johndir. Google are not count all backlinks. Only 65% backlinks are indexed by google. And it's the secret truth.
It's mean all Indexed backlinks are not actually 100% backlinks that's something very strange
Your backlinks are counted when it is indexed. Non-indexed backlinks mean nothing, index backlinks may mean positive or negative depend

Dofollow and Nofollow matters of course it's like you trust a friend and reffering to him or not. so link from me to you say nofollow so i dont credit you or you credit me and rep me.
none backlinks is magic holy grail. and it's a lot of theory and the only way to learn is to read threads wso etc.

If you submit an article into an article directory and a new page was created for your article and it is already indexed by google, than we know that google at least "saw" your link. Would it be added to your blinks profile depends on arcticle directory's quality and google bot's mood. Is the link dofollow or nofollow you can check by viewing the source code of the page.
